Previously Marked Proofread Neurons The objective of this task is to sort through the list of neurons/segments in the sheet and tag them with the appropriately. These neurons are ones that got submitted by mistake or through some other error. To rectify this we will be looking at all of them proofreading and re-tagging them. For cells that are too small or for organelles and dust pieces we need to recategorized as “not a neuron”. For incomplete cells we need to proofread them and re-submit them to the BANC-bot when we are confident they are complete. The complete cells should be labeled “done”. For cells that are too hard to fix or have some errors that make it un-fixable we need to label them as “can’t continue” instead of calling them proofread or complete. You might come across cells that are actually complete. Please mark these with the “already complete” tag. The next slide has step by step instructions.

Review Task -Open sheet -Add your name to the proofreader column G -Use the neuroglancer link in column F to open the task -Right click the segment ID in the link to center it -Proofread the cell, if there is one, and add the appropriate tag in the “Status” Column J Tags: -The "Done" tag is for neurons that weren't complete before that are now "mostly complete" with a high confidence. These should be marked as complete and submitted to BANC-bot -The "not a neuron" tag is for tiny pieces of glia, mitochondria, membranes, extracellular space, dust, myelin, and things of that nature. These should NOT be marked complete with BANC-bot -The "can't continue" tag is for neurons that don't meet the "mostly complete" threshold and should NOT be marked complete with BANC-bot -The "need 2nd opinion" tag is a catch all for cells that don't meet any of the first three criteria and need clarification. Use your colleagues for assistance before adding this tag. -The “already complete” tag should be used after proofreading and you find no errors and the cell is complete without additional edits. These should be re-submitted as complete to BANC-bot -After you finish proofreading a cell, add your updated link to the K column -If your cell is a “need 2nd opinion” then add your link K column as well

What I’ve Found so Far I’ve looked at a few dozen rows so far. They are sorted by size and therefore will have many dust and smaller pieces early on. Towards the end there are larger cells with some issues such as pathswaps and harder areas. There are also a lot of scattered disconnected somas. These should be relatively easy to find the connections for. Good luck and please always ask for help and ask questions so we can better refine our workflows and process. If you see one of my notes but my name isn’t in the proofreader column please still investigate that cell.
